    Diachronic Variation in the Omani Arabic Vernacular of the Al-ʿAwābī District

    In this monograph, Roberta Morano re-examines one of the foundational works of the Omani Arabic dialectology field, Carl Reinhardt’s Ein arabischer Dialekt gesprochen in ‘Oman und Zanzibar (1894). This German-authored work was prolific in shaping our knowledge of Omani Arabic during the twentieth century, until the 1980s when more recent linguistic studies on the Arabic varieties spoken in Oman began to appear. Motivated by an urgent need to expand and reinforce our understanding of Omani Arabic, the book provides a linguistic analysis of the Omani vernacular spoken in the al-ʿAwābī district (northern Oman), based on the speech of fifteen informants recruited throughout the area. It also provides a comparative analysis of the new data with that collected by Carl Reinhardt in 1894. This comparison enables the reader to appreciate the extent of diachronic linguistic variation in the region, and also sheds light on the threats that such variation poses to Omani-specific linguistic features. Organised in four chapters, the book presents a sociolinguistic analysis of the Omani linguistic landscape followed by an examination of the phonology, morphology, and syntax of the al-ʿAwābī vernacular. Each chapter contains primary data collected by the author in situ compared, when applicable, with Carl Reinhardt’s materials. The appendix includes two sample texts extrapolated from the recordings, fourteen proverbs and one traditional song. This study will be of interest to those working in the fields of Omani Arabic, historical and comparative linguistics, translation and interpretation, or those with an interest in how languages develop over time

    Could gestational diabetes mellitus be managed through dietary bioactive compounds? Current knowledge and future perspectives

    Gestational diabetes mellitus (GDM) is a serious problem growing worldwide that needs to be addressed with urgency in consideration of the resulting severe complications for both mother and fetus. Growing evidence indicates that a healthy diet rich in fruit, vegetables, nuts, extra-virgin olive oil and fish has beneficial effects in both the prevention and management of several human diseases and metabolic disorders. In this review, we discuss the latest data concerning the effects of dietary bioactive compounds such as polyphenols and PUFA on the molecular mechanisms regulating glucose homoeostasis. Several studies, mostly based on in vitro and animal models, indicate that dietary polyphenols, mainly flavonoids, positively modulate the insulin signalling pathway by attenuating hyperglycaemia and insulin resistance, reducing inflammatory adipokines, and modifying microRNA (miRNA) profiles. Very few data about the influence of dietary exposure on GDM outcomes are available, although this approach deserves careful consideration. Further investigation, which includes exploring the 'omics' world, is needed to better understand the complex interaction between dietary compounds and GDM

    An Omani Evolving Lexicon: From Carl Reinhardt (1894) to the Present Day

    Oman is a country in constant evolution linguistically, economically and socially. Most of the linguistic studies carried out so far in the Sultanate are located in specific areas of the country and date back to the end of the nineteenth and the beginning of the twentieth century. Carl Reinhardt’s work – Ein arabischer Dialekt gesprochen in ‘Oman und Zanzibar, dated 1894 – is one of the most detailed and richest descriptions of Omani Arabic, specifically of the grammar, including the phonology and morphology, of the Banū Kharūṣ vernacular, spoken in the area of Nizwa and Rustāq (northern Oman), but also among the élite of Zanzibar island. The main purpose of his work was to provide a valuable linguistic guide to the German soldiers quartered on the island and in the Tanganyka region, which were an imperial German colony for a short time. The material supplied by Reinhardt still plays an absolutely essential role for neo-Arabic linguistics and dialectology, although it has some significant issues, such as the lack of Arabic original script and of a comprehensive glossary. Reinhardt’s lexical data, nevertheless, is extremely rich and characterized by some specific traits which make this vernacular different from any other Southern Arabic dialect. In this paper, I will try to outline this richness of Omani lexicon, starting from some examples in Reinhardt’s nineteenth-century core and exploring the variety and changes they underwent over time. These examples will be presented for specific semantic categories (e.g. body parts, food, animals), following the same format as that of the Behnstedt and Woidich's Word Atlas of Arabic dialects / Wortatlas der arabischen Dialekte (2011). Furthermore, a group of specific variations between the original meaning of a root and its different use in the Banū Kharūṣ vernacular, and a few borrowings from foreign languages will be presented and analysed

    Reactive oxygen species derived from NOX3 and NOX5 drive differentiation of human oligodendrocytes

    Reactive oxygen species (ROS) are signaling molecules that mediate stress response, apoptosis, DNA damage, gene expression and differentiation. We report here that differentiation of oligodendrocytes (OLs), the myelin forming cells in the CNS, is driven by ROS. To dissect the OL differentiation pathway, we used the cell line MO3-13, which display the molecular and cellular features of OL precursors. These cells exposed 1–4 days to low levels of H2O2 or to the protein kinase C (PKC) activator, phorbol-12-Myristate-13-Acetate (PMA) increased the expression of specific OL differentiation markers: the specific nuclear factor Olig-2, and Myelin Basic Protein (MBP), which was processed and accumulated selectively in membranes. The induction of differentiation genes was associated with the activation of ERK1-2 and phosphorylation of the nuclear cAMP responsive element binding protein 1 (CREB). PKC mediates ROS-induced differentiation because PKC depletion or bis-indolyl-maleimide (BIM), a PKC inhibitor, reversed the induction of differentiation markers by H2O2. H2O2 and PMA increased the expression of membrane-bound NADPH oxidases, NOX3 and NOX5. Selective depletion of these proteins inhibited differentiation induced by PMA. Furthermore, NOX5 silencing down regulated NOX3 mRNA levels, suggesting that ROS produced by NOX5 up-regulate NOX3 expression. These data unravel an elaborate network of ROS-generating enzymes (NOX5 to NOX3) activated by PKC and necessary for differentiation of OLs. Furthermore, NOX3 and NOX5, as inducers of OL differentiation, represent novel targets for therapies of demyelinating diseases, including multiple sclerosis, associated with impairment of OL differentiation

    An ontology-based approach supporting holistic structural design with the consideration of safety, environmental impact and cost

    Early stage decision-making for structural design critically influences the overall cost and environmental performance of buildings and infrastructure. However, the current approach often fails to consider the multi-perspectives of structural design, such as safety, environmental issues and cost in a comprehensive way. This paper presents a holistic approach based on knowledge processing (ontology) to facilitate a smarter decision-making process for early design stage by informing designers of the environmental impact and cost along with safety considerations. The approach can give a reasoning based quantitative understanding of how the design alternatives using different concrete materials can affect the ultimate overall performance. Embodied CO2 and cost are both considered along with safety criteria as indicative multi-perspectives to demonstrate the novelty of the approach. A case study of a concrete structural frame is used to explain how the proposed method can be used by structural designers when taking multi performance criteria into account. The major contribution of the paper lies on the creation of a holistic knowledge base which links through different knowledge across sectors to enable the structural engineer to come up with much more comprehensive decisions instead of individual single objective targeted delivery

    Extensive molecular profiling of squamous cell anal carcinoma in a phase 2 trial population: Translational analyses of the “CARACAS” study

    Background: Molecular characteristics of squamous cell anal carcinoma (SCAC) are poorly explored. Immune checkpoint inhibitors showed limited activity in phase I/II trials, but predictive and prognostic biomarkers are lacking. Patients and methods: In the phase II randomised trial CARACAS (NCT03944252), avelumab alone (Arm A) or with cetuximab (Arm B) was tested in pre-treated advanced SCAC , with overall response rate being the primary end-point. On pre-treatment tumour tissue samples, we assessed Human papillomavirus status, programmed-death ligand 1 (PD-L1) expression, mismatch repair proteins expression, tumour mutational burden (TMB) and comprehensive genomic profiling by FoundationOne CDx. Tumour-infiltrating lymphocytes were characterised on haematoxylin-eosine-stained samples. Primary objective was to describe response to immunotherapy in the CARACAS trial population according to molecular and histological characteristics. Secondary objectives were to assess progression-free survival (PFS) and overall survival (OS) according to molecular biomarkers. Results: High PD-L1 (>40 with combined positive score) was significantly more frequent in patients with disease control (p = 0.0109). High TMB (>10 mutations per megabase) was related to better OS (hazard ratio (HR) = 0.09; 95%confidence interval (CI) 0.01-0.68; p = 0.019) and PFS (HR = 0.44; 95%CI = 0.15-1.27; p = 0.129). High expression of PD-L1 conferred longer OS (HR = 0.46; 95%CI = 0.19-1.08; p = 0.075) and PFS (HR = 0.42; 95%CI = 0.20-0.92; p = 0.03). Neither OS (HR = 1.30; 95%CI = 0.72-2.36; p = 0.39) or PFS (HR = 1.31; 95%CI = 0.74-2.31; p = 0.357) was affected by high (>1.2) Tumour-infiltrating lymphocytes count. High TMB and PD-L1identified patients were with significantly better OS (HR = 0.33; 95%CI = 0.13-0.81; p = 0.015) and PFS (HR = 0.48; 95%CI = 0.23-1.00; p = 0.015). Conclusions: To our knowledge, TranslaCARACAS is the first study to document prognostic role of TMB and PD-L1 in advanced SCAC patients treated with immune checkpoint inhibitors

    Aims According to cardiovascular outcome trials, some sodium-glucose contransporter-2 inhibitors (SGLT2i) and glucagon-like peptide-1 receptor agonists (GLP-1RA) are recommended for secondary cardiovascular prevention in type 2 diabetes (T2D). In this real-world study, we compared the simultaneous reductions in HbA1c, body weight and systolic blood pressure after initiation of dapagliflozin or GLP-1RA as second or a more advanced line of therapy. Materials and methods DARWIN-T2D was a retrospective multi-centre study conducted at diabetes specialist clinics in Italy that compared T2D patients who initiated dapagliflozin or GLP-1RA (exenatide once weekly or liraglutide). Data were collected at baseline and at the first follow-up visit after 3 to 12 months. The primary endpoint was the proportion of patients achieving a simultaneous reduction in HbA1c, body weight and systolic blood pressure. To reduce confounding, we used multivariable adjustment (MVA) or propensity score matching (PSM). Results Totals of 473 patients initiating dapagliflozin and 336 patients initiating GLP-1RA were included. The two groups differed in age, diabetes duration, HbA1c, weight and concomitant medications. The median follow-up was 6 months in both groups. Using MVA or PSM, the primary endpoint was observed in 30% to 32% of patients, with no difference between groups. Simultaneous reduction of HbA1c, BP and SBP by specific threshold, as well as achievement of final goals, did not differ between groups. GLP-1RA reduced HbA1c by 0.3% more than the reduction achieved with dapagliflozin. Conclusion In routine specialist care, initiation of dapagliflozin can be as effective as initiation of a GLP-1RA for attainment of combined risk factor goals
